Kurzfassung
Das Cytokin Interleukin (IL)-6 besitzt sowohl pro- als auch anti-inflammatorische Eigenschaften und ist ein starker Induktor der Akutphase-Reaktion. Es steht im Zusammenhang mit der Ausbildung von entzündlichen Krankheiten, wie z. B. rheumatoider Arthritis oder Multipler Sklerose. Bei der Therapie von entzündlichen Krankheiten kommen Glucocorticoide, die zur Klasse der Steroidhormone zählen, zum Einsatz, da sie ein breites Spektrum an anti-inflammatorischen Eigenschaften aufweisen. Es ist daher wichtig, die Wechselwirkungen von IL-6 und Glucocorticoiden zu kennen und genauer zu untersuchen. Es ist bereits seit mehreren Jahren bekannt, dass Glucocorticoide die IL-6-induzierte Akutphase-Genexpression in der Leber verstärken. Die zugrunde liegenden molekularen Mechanismen sind jedoch bis heute noch nicht vollständig verstanden. In dieser Arbeit konnte sowohl in der humanen hepatomen Zelllinie HepG2 als auch in primären murinen Hepatozyten gezeigt werden, dass Glucocorticoide die IL-6-induzierte Expression des Feedback-Inhibitors SOCS3 inhibieren, was zu einer Verstärkung der IL-6-induzierten Genexpression führt. Ist die transaktivierende Funktion des Glucocorticoid-Rezeptors gestört, zeigen Glucocorticoide keinen Einfluss mehr auf die IL-6-induzierte Signaltransduktion. In einem zweiten Projekt wurde der Einfluss von Glucagon, einem Antagonisten des Insulins im Körper, auf die IL-6-induzierte Signaltransduktion in primären murinen Hepatozyten untersucht. Es konnte gezeigt werden, dass Glucagon sowohl die IL-6-induzierte Phosphorylierung der MAP-Kinasen ERK1/2 als auch die IL-6-induzierte ERK1/2-abhängige Genexpression inhibiert. Die Regulation der IL-6-induzierten Signaltransduktion durch Glucagon wird über die Aktivierung von PKA und Epac vermittelt, die beide redundant in dem cross-talk zwischen IL-6 und Glucagon involviert sind. Darüber hinaus konnten in Microarray-Analysen neue ERK1/2-abhängige Zielgene von IL-6 identifiziert werden. Zusammenfassend lässt sich sagen, dass in dieser Arbeit ein neuer Wirkmechanismus sowohl von Glucocorticoiden als auch von Glucagon aufgezeigt werden konnte, welcher jeweils die IL-6-induzierte Signaltransduktion weitgreifend beeinflusst.

The cytokine interleukin (IL)-6 exerts pro- and anti-inflammatory properties and is a main inducer of the acute-phase response. It is involved in the pathogenesis of inflammatory diseases like for example rheumatoid arthritis or multiple sclerosis. Glucocorticoids are steroid hormones that are widely used in the therapy of inflammatory diseases because of their anti-inflammatory effects. Therefore, it is important to examine the interactions of IL-6 and glucocorticoids in more detail. It has been known since several years that glucocorticoids increase the IL-6-induced acute-phase gene expression in the liver. However, the underlying molecular mechanisms are still not completely understood. In this work it could be shown both in the human hepatoma cell line HepG2 and in primary murine hepatocytes that glucocorticoids interfere with the IL-6-induced expression of the SOCS3 feedback inhibitor, thereby leading to an upregulation of IL-6-induced gene expression. Impaired transactivation of the glucocorticoid receptor abolishes the effect of glucocorticoids on IL-6-induced signal transduction. In a second project the influence of glucagon, an antagonist of insulin in the body, on the IL-6-induced signal transduction was investigated in primary murine hepatocytes. It could be shown that glucagon inhibits the IL-6-induced phosphorylation of the MAP kinases ERK1/2 and the IL-6-induced ERK1/2-dependent gene expression by redundant action of PKA and Epac. Moreover, in microarray analyses new ERK1/2-dependent target genes of IL-6 could be identified. In sum, a new mechanism of both glucocorticoid and glucagon action, each with a broad effect on IL-6 signaling, could be revealed in this study.
