Dephosphoryliertes Fetuin-A zeigte in beiden Assays eine reduzierte Aktivität, „hyper“phosphoryliertes Fetuin-A gesteigerte Aktivität. Fetuin-A is a multifunctional phospho-glycoprotein. One of its central roles is to bind calcium phosphates in the blood and facilitate their transport. Although it is known that fetuin-A exists in various phosphorylation states in the circulation, it remained unclear whether this post-translational modification affected its mineral-binding capacity. For other mineral-binding proteins, such as osteopontin or matrix Gla protein, such a functional link has already been described. The aim of this study was to investigate whether the phosphorylation status of fetuin-A influenced its ability to bind mineral. To this end, recombinant fetuin-A was expressed in ExpiCHO-S™ cells and in vitro phosphorylated by a self-engineered, active variant of the extra-cellular kinase Fam20C, or in vitro dephosphorylated using alkaline phosphatase. The phosphorylation status was confirmed by Phostag™ SDS-PAGE and protein mass spectrometry. Functional analysis was performed using two in-house developed assays: the calcium phos-phate precipitation inhibition - CAPPI assay, which measures the ability of a reagent to stabi-lize saturated calcium (5 mM) and phosphate (3 mM) solutions, and the T50 assay, which quantifies the propensity of a solution to undergo calcification. The results clearly demonstrate that phosphorylation regulates the fetuin-A mineral-binding function. Dephosphorylated fetuin-A showed reduced, and in-vitro “hyper”phosphorylated fetuin-A had increased capacity to inhibit calcium phosphate precipitation in both assays. This study thus provides the first direct functional evidence that the phosphorylation status of fetuin-A critically regulated its mineral-binding behavior. These findings have important implications for understanding physiological calcification processes and may inform the development of therapeutic strategies involving modified or recombinant forms of fetuin-A.
