Kurzfassung
Ziel des ersten Teils dieser Doktorarbeit war die nicht enzymatische Synthese des 7ß-N-Acetyglucosaminides der UDCA. Dieses Konjugat sollte für in vitro-Untersuchungen zum Wirkmechanismus der UDCA dienen. Die Synthese war sehr aufwendig und schwierig, die Ausbeute gering. Ziel des zweiten Teils dieser Arbeit war den Metabolismus von Isoursodeoxycholsäure beim Menschen zu untersuchen. IsoUDCA wurde mit >99% Reinheit synthetisiert und für eine Woche, 3x250 mg täglich, sechs gesunden männlichen Probanden verabreicht. Die Gallensäuren wurden jeweils vor und nach der Einnahme aus Duodenalgalle, Serum und 24-h Sammelurin extrahiert, in die verschiedenen Konjugate getrennt und mittels Gaschro-matographie-Massenspektrometrie und Fast Atom Bombardment-Massenspektrometrie analysiert. Ergebnisse dieser Studie mit IsoUDCA zeigen zum einen die gute Resorption und die hervorragende Verträglichkeit dieser Substanz und zum anderen der biochemische Befund der fast vollständigen hepatischen Umwandlung von IsoUDCA zu UDCA.

The aim of the first part of this thesis was the non-enzymatic synthesis of 7ß-N-acetyglucosaminides of UDCA. This conjugate should serve for in vitro investigations of the mechanisms of UDCA. The synthesis was very complex and difficult, and the yield was quite small. The aim of the second part of the study was to examine the metabolism of isoursodeoxycholic acid (isoUDCA) in humans. IsoUDCA was synthesized with >99% purity> It was administered orally to six healthy males for 1 week (3x250 mg daily). Bile acid were extracted from duodenal bile, serum, and 24-h urine samples collected before and at the end of the study period, separated into distinct conjugates, and subsequently analysed by gas chromatography mass spectrometry as well as fast atom bombardment mass spectrometry. This study demonstrates good tolerance and significant intestinal absorption of orally administered isoUDCA. IsoUDCA is extensively isomerised, probably both by intestinal and hepatic enzymes to yield UDCA, which becomes the major biliary compound.
