Kurzfassung
Das Plasmaprotein alpha-2-HS-Glycoprotein(AHSG)/Fetuin inhibiert die Calciumphosphat-Präzipitation aus übersättigter Lösung transient. Durch Struktur-Funktionsuntersuchungen wurde das inhibitorische Strukturmotiv innerhalb der ersten Cystatin-ähnlichen Domäne von AHSG/Fetuin eingegrenzt und durch Vergleiche mit weiteren Domänen der Cystatin-Superfamilie (Fetuin-B (FETUB), Kininogen (KNG) und Histidine Rich Glycoprotein (HRG)) näher charakterisiert. Da von diesen Domänen keine Strukturdaten verfügbar sind, wurden Modelle durch komparative Strukturmodellierung erstellt. Das Modell der murinen AHSG Domäne D1 zeigt eine aminoterminale alpha-Helix, um die sich ein vier-strängiges beta-Faltblatt wölbt. Auf dem beta -Faltblatt bilden sieben saure Aminosäure-Seitenketten einen zusammenhängend negativ geladenen Oberflächenbereich. Keine andere Cystatin-ähnliche Domäne in FETUB, KNG oder HRG besitzt einen vergleichbaren negativen Ladungsbereich, was zu der Annahme führte, daß die Inhibition über eine Interaktion dieser sauren Seitenketten mit den Calcium-Ionen der Mineraloberfläche erfolgt. Der Test des inhibitorischen Potenzials rekombinant exprimierter Fragmente der murinen AHSG Domäne D1 offenbarte, daß die von den Aminosäuren 15-70 gebildete Struktur, d.h. die aminoterminale alpha -Helix und die ersten beiden Stränge des darauffolgenden beta -Faltblattes, bereits das komplette inhibitorische Potenzial beinhaltet. Weiterhin zeigten die Inhibitionstests, daß keine andere Cystatin-ähnliche Domäne einen vergleichbar guten Inhibitor darstellt, da ihnen, wie die Strukturmodelle zeigten, eine entsprechende Anhäufung saurer Seitenketten auf dem beta -Faltblatt fehlt. Durch Test des verbleibenden inhibitorischen Potenzials in den Seren von Dialyse-Patienten und von Ahsg-defizienten Mäusen mit teilweise massiven ektopischen Calcifizierungen konnte die klinische Relevanz von Fetuin als wichtigem löslichen Inhibitor belegt werden. Bei Betrachtung des Löslichkeitsproduktes von Hydroxylapatit und der maximal von AHSG gebundenen Calcium Ionen wird deutlich, daß die Inhibition nicht einfach auf einer Erniedrigung des Ionenproduktes durch Bindung von Calcium-Ionen an die sauren Seitenketten beruhen kann. Die Inhibition der Calciumphosphat-Präzipitation beruht vielmehr auf der Interaktion von AHSG mit Calciumphosphat-Oberflächen. Zunächst wurde der Einfluss von Fetuin auf die Morphologie des Präzipitates mittels Rasterelektronenmikroskopie untersucht. Danach gelang der Nachweis löslicher Calciumphosphat-Fetuin Aggregate mittels Transmissionselektronenmikroskopie. Fetuin bindet mit hoher Affinität an die Oberfläche von Kristallisationskeimen und wirkt somit auf deren Wachstum, durch die erniedrigte Diffusionsrate von Calcium- und Phosphat-Ionen an die Oberfläche, verlangsamend.

The plasma protein alpha-2-HS-glycoprotein (AHSG)/fetuin transiently inhibits the calcium phosphate precipitation from supersaturated solution. The unique structural motif within the first domain of AHSG mediating the inhibition of precipitation was studied by use of structure-function relations of protein domains of the cystatin superfamily. Since no structural data of these cystatin-like domains were available, models were generated by comparative structure modelling. The model of AHSG domain D1 shows an aminoterminal alpha-helix and a bent four-stranded beta-sheet. Seven acidic amino acids form a contiguous and negatively charged surface area. No other cystatin-like domain contains a comparable negatively charged surface area, suggesting that the inhibition is mediated by an interaction of these acidic residues with calcium ions on the mineral surface. The effiency of recombinantly expressed deletion mutants of the murine AHSG domain D1 revealed that the structure built up by the amino acids 15-70, i.e. the aminoterminal alpha-helix and the first two strands of the beta sheet, contain the complete inhibitory potential. Futhermore, functional testings evidenced that no other cystatin-like domain inhibits the precipitation with a comparable effiency because their modelled structures lack a comparable density of acidic residues on the beta-sheet. Tests of the remaining inhibitory potential of sera from dialysis-patients and Ahsg deficient mice that both already had developed severe ectopic calcifications in various organs, proved the importance of AHSG as an essential soluble inhibitor of calcium phosphate precipitation. Considering the solubility product of hydroxyapatite and the maximal amount of AHSG bound calcium, the inhibition cannot be explained by a reduction of the ion product. In fact, the inhibition relies on the adsorption of AHSG on calcium phosphate surfaces. This was demonstrated firstly by a change in morphology of the precipitate induced by AHSG as observed by scanning electron microscopy and secondly by visualisation of soluble AHSG-calcium phosphate aggregates by transmission electron microscopy. Obviously, AHSG binds to nascent calcium phosphate aggregates with a high affinity and thus prevents their further growth by a reduction of the diffusion rate of calcium and phosphate ions to the surface.
