Kurzfassung
Das Mikrozirkulationsnetzwerk der Chorioallantois-Membran (CAM) wird in vielen Bereichen der Forschung als Angiogenese-Modell verwendet. Bisher war die Auswertung des Gefäßsystems nicht zufriedenstellend, da sie durch subjektive Klassifizierung ausgeführt wurde und die kleinsten Gefäße, die Kapillaren, nicht erfasst werden konnten. Zur Gefäßdetektion wurde ein neuer Weg eingeschlagen, indem die transparenten Kapillaren mit Hilfe der Erythrozytenbewegungen detektiert werden, ohne dass ein invasiver Eingriff notwendig ist. Die Erythrozytenbewegungen werden unter Berücksichtigung ihrer Absorptionsbande in einem digitalen Film aufgezeichnet und analysiert. Dabei wurde die Bewegung der gesamten Membran von der Bewegung der Erythrozyten getrennt. Die Detektion der transparenten Kapillargefäße wurde mit einer Histogrammanalyse durchgeführt. Das Resultat ist ein Binärbild des Gefäßbaumes. Zur Analyse des Gefäßbaumes wurde ein Wellenalgorithmus entwickelt, der das Gefäßnetz wie eine Welle erfasst. Hiermit ist es möglich, den Einfluss des Rausches im Bild zu reduzieren. Weiterhin ist dieser Algorithmus geeignet, Bifurkationen, Gefäßdurchmesser und Gefäßabschnitte zu bestimmen. Der Einfluss von Biomaterialien, die als Bauchdeckenersatz in der Chirurgie verwendet werden, wurde mit Hilfe der fraktalen Dimension quantifiziert. Hierbei konnte erstmals gezeigt werden, dass die fraktale Dimension der CAM abhängig ist vom Abstand zum Biomaterial. Mit der Detektion ist eine objektive Beurteilung der Gefäßstruktur der CAM möglich. Die Methode kann übertragen werden auf Untersuchungen zur Angiogenese der CAM bei Tumorapplikationen oder auf andere angiogenetische Tests, die auf der CAM durchgeführt werden.The microvascular network of the chorioallantoic membrane (CAM) is frequently used as a model to investigate angiogenesis. Up to now, the quantification of the vessel system has been unsatisfactory because it was performed by a subjective classification without regard to the capillaries. For the detection of the transparent capillaries the movement of the erythrocytes was used in a non-invasive process. To this end, the erythrocyte flow was recorded in a digital film sequence. To detect the capillaries the movement of the membrane was separated from the erythrocyte movement. The capillaries were separated from the background by analyzing a histogram. The result is a binary image of the microvascular network. A wave-like algorithm was established to extract the interesting parameters of the vessel system . The influence of noise can be reduced by this algorithm. Also the numbers of bifurcations, the cross section of the vessels and the length between two bifurcations can be evaluated. The influence of biomaterials, like prosthetic meshes for hernia repair, was quantified by the fractal dimension. It was shown that the fractal dimension is a function of the distance to the biomaterial. With this new method the microvascular network of the CAM can be evaluated automatically. It can also be used for all angiogenetic tests on the CAM, such as the investigation of tumor tissue.
