Kurzfassung
Der Transkriptionsfaktor Mad1 ist ein potentieller Tumorsuppressor. Innerhalb des Myc/Max/Mad-Netzwerks antagonisiert es die transformierenden Eigenschaften des Proto- Onkogens c-Myc. Es ist beschrieben worden, dass Mad1 die Transformation von REFs durch c-Myc/Ras verhindern kann, indem es den G1/S-Übertritt blockiert. Ausserdem konnte gezeigt werden, dass Mad1 ein Inhibitor der Apoptose ist. Die Mitglieder des Myc/Max/Mad- Netzwerkes sind Transkriptionsfaktoren und regulieren Gen-Expression nach Bindung an spezifische DNA-Sequenzen über Rekrutierung chromatin-modifizierender Faktoren. Prinzipiell gilt c-Myc als Aktivator und Mad1 als Repressor der Transkription. In der Entwicklung myeloider Zellen zeigte sich Mad1 als essentieller Faktor, dessen (funktioneller) Verlust in einer stark eingeschränkten und/oder verzögerten Differenzierung von Granulozyten und Monozyten resultiert. Dazu passt der Befund, dass mad1 in der TPA- und G-CSF-induzierten Differenzierung humaner monozytärer U937-Zellen hochreguliert wird. Es ist deshalb interessant zu erfahren, welche anderen Transkriptionsfaktoren und welche intrazellulären Signaltransduktionswege an der Regulation des mad1-Gens beteiligt sind. So lassen sich zentrale zellbiologische Prozesse wie Proliferation und Differenzierung besser verstehen. Im Rahmen dieser Arbeit konnte ein DNA-Abschnitt mit Promotor-Funktion und wichtigen regulatorischen Elementen identifiziert werden. Dieses Promotor-Fragment liegt wenige Basenpaare oberhalb des Translationsstarts, zeigt im Alignement mit der korrelierenden Sequenz von Maus und Ratte eine fast 70%ige Homologie und enthält zahlreiche Bindestellen für Transkriptionsfaktoren, deren Relevanz in der myeloiden Differenzierung in mehreren Untersuchungen gezeigt werden konnte. Der mad1-Promotor ist sehr GC-reich und enthält keine TATA-Box. Es konnte ein Transkriptionsstart des mad1-Gens bestimmt werden. Dieser befindet sich in der homologen Promotor-Region am 3´-Ende. Der Einfluss des Chromatins und seiner Modifikationen auf die Transkription ist bekannt. Deshalb ist es wichtig die nukleosomale Organisation in regulatorischen DNA-Bereichen genauer zu untersuchen. Nach dem nucleosomal mapping des mad1-Promotors und des ersten Exons zeigte sich, dass die zuvor identifizierte homologe Region nukleosomenfrei ist. Das passt gut zu anderen Befunden, die nahelegen, dass eine uneingeschränkte Zugänglichkeit zum Promotor eine Voraussetzung der Transkriptionsinitiation ist. Leider zeigten sich keine G-CSF- und TPA-abhängigen Veränderungen der nukleosomalen Struktur in diesem Bereich des mad1-Gens. Die Homologie-Box des mad1-Promotors ist sowohl in proliferierenden als auch in differenzierenden Zellen nukleosomenfrei. Um den Einfluss verschiedener Transkriptionsfaktoren auf die mad1-Expression im Detail zu untersuchen, wurde der humane mad1-Promotor als Reportergen-Konstrukt kloniert und es wurden von ihm Deletionsmutanten hergestellt. In darauffolgenden Reportergen-Assays konnten c-Myb, PU.1 und C/EBP Alpha, Beta und Epsilon als potente Transaktivatoren nachgewiesen werden. Im Falle von c-Myb zeigte sich darüber hinaus eine eindeutige G-CSF-Abhängigkeit. Die vornehmlich durch den G-CSF/R aktivierten STAT1 und STAT3 weisen aber nur ein geringes Transaktivierungspotential auf. Mit Hilfe der Chromatinimmunpräzipitation ist es möglich sehr spezifisch und sehr sensitiv Protein-DNA-Interaktionen in vivo zu untersuchen. So konnte am humanen mad1-Promotor die Bindung der RNA-Polymerase-II und des Transkriptionsfaktors PU.1 nachgewiesen werden, somit Befunde aus in vitro-Experimenten teilweise bestätigt werden. Zusätzlich sind die Histone im Promotorbereich acetyliert, deren ChIP-Signal steigt erst im Bereich der Homologie-Box an und reicht bis ins 1.Exon hinein. Für die anderen in vitro getesteten Faktoren (STAT1, STAT3, C/EBPe) wurde nach zwei Stunden G-CSF-Stimulation keine Bindung an den mad1-Promotor nachgewiesen. Auch zeigte sich in den ChIP-Experimenten keine G-CSF-abhängige Rekrutierung von Transkriptionsfaktoren. Diese Ergebnisse belegen eindeutig die essentielle Rolle, die die identifizierte DNA-Region in der Regulation der mad1-Expression spielt und weisen in diesen Prozess involvierte Transkriptionsfaktoren nach. Diese Arbeiten werden im Rahmen einer naturwissentschaftlichen Doktorarbeit fortgeführt. Hier gilt es, in Reportergen-Assays die genauen Bindestellen für oben genannte Transkriptionsfaktoren zu definieren und deren Kooperativität am mad1-Promotor zu untersuchen. Ausserdem ist es wichtig, die mad1- Expression in einen G-CSF-kontrollierten Kontext zu bringen. Mit Hilfe der Chromatinimmunpräzipitation sollen in vivo intrazelluläre Mechanismen untersucht werden, die die Bindung von Transkriptionsfaktoren an den mad1-Promotor und dessen Expression regulieren. Auch chromatin-modifizierende Prozesse bleiben weiterhin interessant im Hinblick auf die transkriptionelle mad1-Regulation.The transcriptionfactor Mad1 is a potential tumorsuppressor. As a member of the Myc/Max/Madnetwork it antagonizes the transforming ability of the protooncogene c-myc. It was shown, that Mad1 can inhibit the transformation of REFs by c-myc/Ras through blockade of the G1/S-transition. Moreover it was shown, that Mad1 is an inhibitor of apoptosis. The members of the Myc/Max/Madnetwork are transcriptonfactors and regulate gene-expression after binding to specific DNA-sequences and then recruiting chromatin-modifying factors. In principle c-myc is described as an activator of transcription while mad1 is described as an inhibitor of transcription. Mad1 is an essential factor in the development of myeloid cells and loss of Mad1-function results in delayed differentiation of granulocytes and monocytes. Accordingly to this it fits, that mad1 gets upregulated after TPA- or GCSF-stimulation of U937-cells. Therefore it is interesting to know how Mad1 itself is transcriptionally regulated, to get more insight into crucial cellular processes like proliferation and differentiation. What are essential extracellular factors and what different intracellular signaltransduction-pathways are involved? In this study a human DNA-fragment was isolated which has mad1-promoter-function and contains a set of regulatory elements like binding sites for transcriptionfactors. This promoter-fragment is located only a few basepairs upstream of the translation-start-site. Alignement of this human sequence with DNA-sequences of mouse and rat reveales a box with nearly 70% homology and binding sites for transcriptionfactors important for myeloid development and differentiation. The mad1-promoter is very GC-rich and contains no TATA-box. In this study one transcription-start-site of the mad1-gene was identified. It is located at the 3´-end of the homologous promoter region. The influence of chromatin and its modifications on transcription is known. So it is important to investigate the nucleosomal structure of cis-acting DNA-elements like promoters and/or enhancers. In this study nucleosomal mapping of the mad1-promoter in U937-cells was done. It reveales that exactly the previous identified homologous region is nucleosome-free. This fits to other results, which show, that free access to regulatory DNA is a prereqiusite for transcription initiation. Unfortunately no GCSF- or TPA-effect on the nucleosomal structure was observed. The homology-box of the mad1- promoter is nucleosome-free in proliferating and/or differentiating U937-cells. To investigate the influence of different transcriptionfactors on Mad1-expression in detail, the Mad1- promoter was cloned as a reportergene-construct and several deletion-mutants were constructed. In following reportergene-assays c-myb, PU.1 and C/EBP Alpha, Beta and Epsilon were identified as positive regulators of mad1-transcription in RK13-cells in vitro. In the case of c-myb, cotransfection of GCSF/R and stimulation with GCSF results in a twofold increase of luciferase-activity. STAT1 and STAT3 exhibit only low but GCSF-dependent transcriptional potential. The ChIP-method (Chromatin-Immunpräzipitation) is very sensitive and specific in investigating protein/DNA-interactions in vivo. Using ChIP binding of RNA-Pol II and PU.1 to the human mad1- promoter was detected. This confirms some in vitro results. Additionally with the ChIP-method also histone-acetylation at the 3´-side of the homology-box was observed. But for other transcriptionfactors like STATs or C/EBPs no DNA-binding to the mad1-promoter was detected. Also no GCSFdependent recruitment of factors or histone-modification. In conclusion the results obtained in this study prove the essential role of the identified DNA-region in regulation of mad1-transcription and reveales the nucleosomal structure of this mad1-promoter. Also potential transcriptionfactors involved in upregulating mad1-expression were detected. But to completely understand the regulation of mad1-expression further investigation like timedependent and cofactor-dependent ChIP- and Reportergene-assays are needed.
