Kurzfassung
Katalyse in einem invertierten Zweiphasensystem scCO2/H2O, bei dem der Katalysator in der stationären scCO2-Phase immobilisiert ist und Substrat und Produkt in der wässrigen Phase gelöst sind, stellt eine interessante Alternative zu klassischen Zweiphasensystemen dar und ermöglicht insbesondere die Umsetzung polarer Substrate unter Verwendung von CO2 als Lösungsmittel. Da es viele Beispiele für hydrophile Feinchemikalien und biologisch aktive Substanzen gibt, liefert eine Methode, diese Verbindungen ohne die Probleme der Produktisolierung und Katalysatorrezyklisierung synthetisieren zu können, eine attraktive Möglichkeit. Aus Sicht der Verfahrenstechnik hat ein invertiertes System den Vorteil, dass die stationäre CO2-Phase, die den Katalysator enthält, während des Prozesses den Reaktor nicht verlässt und daher nicht entspannt und wieder komprimiert werden muss. Auf diese Weise entfällt ein energie- und kostenintensiver Schritt. Das Ziel der vorliegenden Arbeit war es, die Grundlagen der Anwendung eines invertierten Zweiphasensystems scCO2/H2O zu untersuchen. Im Anschluss an die Bereitstellung eines geeigneten Katalysatorsystems und den Aufbau der Versuchsanlage wurde die prinzipielle Anwendbarkeit dieses Systems sowie die Rezyklisierbarkeit des Katalysators am Beispiel der Hydrierung von Itaconsäure als Modelsubstrat gezeigt. Verschieden „CO2-phile” Rhodiumkomplexe wurden als Katalysatoren eingesetzt. Die besten Ergebnisse zeigten sich bei Verwendung des fluorierten Phosphanliganden 3-H2F6TPP und [Rh(cod)2]BF4. Bei einem Rhodium:Ligand:Substrat-Verhältnis von 1:5:200 erreicht man nach einer Stunde Reaktionszeit einen Umsatz von 95%. Unter den gewählten Bedingungen konnte der Katalysator dreimal rezyklisiert werden. Man erhält eine Umsatzzahl von 800, wobei das Ende der Katalysatoraktivität noch nicht erreicht wurde. Anschließend erfolgten die Optimierung der Reaktionsparameter, die Hydrierung weiterer polarer Substrate sowie die Durchführung asymmetrischer Hydrierungen in diesem System. Letztere lieferte mit (R,S)-3-H2F6-BINAPHOS-modifizierten Rhodiumkomplexen exzellente Ergebnisse. Die Hydrierung von Itaconsäure gelang bei vollständigem Umsatz und einem Enantiomerenüberschuss von 97-98% ee (S) und auch die Rezyklisierung des Katalysators war erfolgreich. Die Hydrierung von Methyl-2-acetamidoacrylat lieferte über fünf Zyklen hinweg vollständigen Umsatz und hohe Enantioselektivität von > 98% ee (R). In einem weiteren Teilprojekt wurden verschiedene Materialien und Substanzklassen auf ihre Löslichkeit in scCO2 untersucht, da die Löslichkeit des Katalysators in scCO2 entscheidend für die Anwendbarkeit des invertierten Zweiphasensystems scCO2/H2O ist. Dazu wurde eine einfache Methode zur Abschätzung von Löslichkeiten verschiedener Ligandensysteme, die mit sehr geringen Substanzmengen auskommt, entwickelt und angewendet.

Catalysis using an inverted scCO2/H2O biphasic system where the catalyst is immobilized in scCO2 as stationary phase and the substrate and product are contained in the aqueous phase, offers an interesting alternative to classical biphasic systems and enables the conversion of polar substrates, using scCO2 as solvent. As many fine chemicals and biological active compounds are hydrophilic in nature, a method for synthesizing these products without the problems of isolation and catalyst recycling forms an attractive possibility. The use of scCO2 as solvent reduces gas-liquid-liquid mass transfer limitations and eliminates cross-contamination of organic solvents into the aqueous layer. The catalyst is easily recycled without depressurising the autoclave by removal of the aqueous layer via a needle valve followed by addition of the next batch of substrate using a HPLC pump. The aim of this thesis was to examine the application of an inverted scCO2/H2O biphasic system. After allocation of an applicable catalyst and set-up of the reactor, the general practice of the system and the ability to recycle the catalyst was tested in case of the hydrogenation of itaconic acid as model substrate. Several CO2-philic rhodium-complexes were used. The best results were achieved using the fluorinated phosphine ligand 3-H2F6TPP and [Rh(cod)2]BF4. With a rodium:ligand:substrate ratio of 1:5:200, a conversion of 95% after 1 h reaction time was reached. Under the chosen conditions, the catalyst could be recycled at least three times. A turnover number of 800 was reached. After this, the optimization of reaction parameters occurred and other polar substrates were used. Then, enantioselective Hydrogenation was accomplished, leading to excellent results when (R,S)-3-H2F6-BINAPHOS-modified rhodium complexes were used. Hydrogenation of itaconic acid led to an enantiomeric excess of 97-98% ee (S) at full conversion and with recycling of the catalyst. Hydrogenation of Methyl-2-acetamidoacrylate gave full conversion and an enantiomeric excess of >98% ee (R) for at least five cycles. Another subject was the determination of solubility in scCO2, as the catalyst solubility plays an important role for the application of the inverted scCO2/H2O biphasic system. Therefore a simple method for qualitative estimation of the solubility, requiring only small amounts of compound was developed and applied on different materials and substances.
