Although a function of haptoglobin on macrophage adherence has been proposed (Sharpe-Timms 2002), the direct mechanism has not been investigated. Up to now limited information on the amount of haptoglobin in uterine secretions depending on the cyclic stage are available. Methods: The amount of haptoglobin from ovulation to menstruation in uterine secretions has been determined by quantitative Western blotting. Furthermore the influence of 0, 10, 50, 100 µg/ml haptoglobin1-1 or 2-2 on the adhesion of CD11b positive leukocytes to an extracelluar matrix was investigated. Results: At day 18/19 of the cycle lowest levels of haptoglobin in uterine secretions where detected (12.9 ng/5µg protein; ß-haptoglobin). This was followed by a significant increase at periimplantation time to highest levels measured (26.9 ng/5µg protein; ß-haptoglobin). Both, haptoglobin1-1- and 2-2 significantly reduced the binding of CD11b positive leukocytes to extracelluar matrix. Nevertheless, haptoglobin1-1 was more effective than 2-2. Conclusion: Haptoglobin is reducing the binding of leukocytes to extracellular matrices via CD11b receptor. In uterine secretion during secretion phase about 130 to 220 ng/µl haptoglobin are detectable. This amounts are highly effective to reduce leukocyte binding to ECMs.</dc:description><dc:source>Aachen : Publikationsserver der RWTH Aachen University 58 S. : Ill., graph. Darst.
