Penetrationsfördernde polymere Mizellen für die Behandlung fibrotischer Tumore

Note: Fibrotische Tumore weisen eine schlechte Prognose auf und sind sehr schwer behandelbar. Das fibrotische Tumorstroma zeichnet sich durch seine besondere Dichte und eine Dysregulation der extrazellulären Matrix mit übermäßiger Kollagenablagerung aus, was die Akkumulation, Penetration und Distribution von (Nano)Wirkstoffen in Tumoren und Metastasen kompliziert. Um diese Barrieren zu überwinden und die Therapie fibrotischer Tumore zu verbessern, setzen wir auf einen kombinatorischen Ansatz mit doppelt beladenen Mizellen, welche sowohl ein Chemotherapeutikum als auch einen Modulator des Tumor Mikromilieus beinhalten. Konkrete Ziele sind: (1) Entwicklung π-π-stabilisierter polymerer Mizellen mit einer Größe von 30-40 nm und einer sehr hohen Beladungsdichte; (2) Evaluierung des therapeutischen und des „Priming“ Potentials von Paclitaxel und Dexamethason beladenen Mizellen im Hinblick auf Reduktion der Tumorzelldichte und der extrazellulären Matrix; (3) Analyse des Effekts von Tumor-Priming mit doppelt beladenen Mizellen in Bezug auf ihre Tumorakkumulation, Penetration und Verteilung; und (4) Eruierung der therapeutischen Effizienz der doppelt beladenen Mizellen als Monotherapie sowie in klinisch relevanten Kombinationstherapien. Das Potential der Mizellen wird in zwei subkutanen Tumor-Mausmodellen (BxPC3 Pankreas- und 4T1 Mammakarzinom), im spontanen und orthotopen KPC Pankreaskarzinom, und in Tumore stammend von Brustkrebs-Patientinnen (patient-derived xenografts) untersucht. Multimodale und multiskalige Bildgebungsverfahren werden zur Analyse der Akkumulation, Penetration, Distribution und Effizienz der doppelt beladenen Mizellen eingesetzt. Dieses Projekt wird dazu beitragen, ein besseres Verständnis der biologischen Barrieren im tumorgerichteten Wirkstofftransport zu erlangen, sowie neuartige chemische und pharmazeutische Technologien zu etablieren, um die Therapie fibrotischer Tumore zu verbessern.
