Kurzfassung
Proteasen der ADAM-Proteinfamilie spalten verschiedenste Oberflächenproteine. Dadurch haben sie entscheidende Funktionen in der Embryogenese, im Entwicklungsprozess, in der Entstehung von Krebs und im Schutz vor neurodegenrativen Krankheiten. ADAM-Proteasen sind weiterhin auf allen Ebenen des Entzündungsrozesses beteiligt: Von der Cytokinfreisetzung während der entzündlichen Aktivierung, über die Leukozytenrekrutierung bis hin zur Regeneration des Gewebes. Die transendotheliale Migration von Leukozyten ist ein entscheidender Schritt im Entzündungsgeschehen. Dieser Migrationprozess beruht auf dem fein abgestimmten Zusammenspiel von Endothelzellen und Leukozyten. Mehrere Studien zeigten, dass im Entzündungsgeschehen endotheliales ADAM10 und 17 die endotheliale Permeabilität und dadurch die Migration von Leukozyten beeinflussen. In vivo-Studien zeigten die Bedeutung von leukozytär exprimiertem ADAM8, 10 und 17 in der Diapedese von Leukozyten. Obwohl leukozytäres ADAM10 von zentraler Bedeutung in der lymphoiden Reifung ist, besteht immer noch Unklarheit über seine Rolle in der Rekrutierung von Leukozyten. In der vorliegenden Arbeit zeigte sich, dass die pharmakologische Inhibition von ADAM10 zu einer Reduktion der transendothelialen Migration und der Chemotaxis der monozytischen Zelllinie THP1 führte. Um diese Befunde zu bestätigen, wurde ein lentivirales shRNA-System etabliert, um ADAM8, 9, 10 und 17 transskriptionell zu inhibieren. Im Vergleich von THP1- Zelllinien mit einer Herunterregulation von ADAM8, 9, 10 oder 17 in transendothelialer Migration und Chemotaxis zeigte sich, dass ADAM8 und 10 von kritischer Bedeutung für die Migration von THP1-Zellen sind. Im Gegensatz hierzu zeigte die Herunterregulation von ADAM9 und 17 keinen bzw. nur einen geringen Einfluss auf die Migration von THP1-Zellen. Folgeexperimente zeigten, dass weder die Sensivität von THP1-Zellen gegenüber dem chemotaktischen Stimulus MCP-1 noch ihre Calcium-Antwort nach MCP-1 Stimulation von der transskriptionellen Inhibierung der ADAM-Proteasen beeinflusst wurde. Ebensowenig war die Adhäsion von THP1-Zellen gegenüber VCAM-1, ICAM-1 und Fibronektin von der pharmakologischen Inhibierung oder der transskriptionellen Herunterregulation der untersuchten Proteasen betroffen. Dies ist die erste Studie, die die vier Proteasen ADAM8, 9, 10 und 17 im Migrationsgeschehen von Leukozyten analysiert und direkt vergleicht. Die Studie zeigt, dass ADAM8 und 10 eine entscheidende Rolle in der in vitro-Migration und Chemotaxis der monozytischen Zelllinie THP-1 spielen. Hierbei bietet sie die Grundlage für weitere Studien über den molekularen Mechanismus, über den ADAM8 und 10 an der Migration von Leukozyten beteiligt sind.

Proteases of the ADAM family are involved in the proteolytic shedding of various cell surface molecules. Thereby, they fulfil essential functions in embryogenesis, development, neuro-protection and cancer development. ADAM proteases also participate in all stages of inflammation, starting from early-response cytokine release and leukocyte recruitment to the process of tissue repair and damaged tissue revascularisation. The transendothelial migration of leukocytes from the blood into the inflamed tissue is an important hallmark of inflammatory responses to infections or trauma. This migratory process is dependent on well-orchestrated responses of both endothelial and leukocytic cells. Endothelial ADAM10 and 17 have been shown to regulate endothelial permeability and thereby leukocyte transendothelial migration in inflammatory settings. In vivo studies demonstrated the importance of leukocytic-expressed ADAM8 and ADAM17 in the recruitment of leukocytes. While ADAM10 is important in the development of lymphoid cells, its role on leukocytes during transendothelial migration has yet to be fully elucidated. The use of pharmaceutical ADAM inhibitors in in vitro transendothelial migration experiments revealed that transendothelial migration and the chemotaxis of THP1 cells was inhibited in the presence of an ADAM10 specific inhibitor. To confirm these findings a lentiviral shRNA system for the transcriptional inhibition of ADAM8, 9, 10 and 17 was established. The comparison of THP1 cells with a gene silencing of ADAM8, 9, 10 or 17 in transendothelial migration and chemotaxis experiments revealed that ADAM8 and ADAM10 are critically involved in THP1 cell migration. By contrast, ADAM9 expression did not seem to be required and ADAM17 was of minor importance for the migration of THP1 cells. Subsequent experiments showed that neither the sensitivity of THP1 cells towards the chemotactic stimulus MCP-1 nor their Calcium-response to MCP-1 was affected by the gene silencing of the investigated ADAM proteases. Neither was the adhesion of THP1 cells to the recombinant integrin ligands VCAM-1, ICAM-1 and fibronectin affected by the pharmaceutical inhibition or the gene silencing of the investigated proteases. This is the first study to analyse and directly compare the role of the four proteases ADAM8, 9, 10 and 17 in the migration of leukocytic cells. It shows that ADAM8 and ADAM10 are crucial for in vitro transendothelial migration and chemotaxis of the monocytic cell line THP1. This study has identified the previously unknown importance of ADAM8 and ADAM10 in cell migration and therefore provides the basis for further analysis of the molecular mechanism by which ADAM8 and ADAM10 contribute to leukocyte migration.
