Kurzfassung
The first work “The copper-catalyzed Sonogashira-Hagihara-type cross-coupling reactions” described a sub-mol% copper-catalyzed system for the cross-coupling reactions of aryl halides and terminal alkynes. In the process, all the tested copper salts were effective and it was determined that the structure of the ligand was crucial for the success of reactions. In addition, the reaction scope and mechanistic details by kinetic measurements and DFT (density functional theory) were investigated. The second work “Copper-mediated direct oxidative cross-couplings of 1,3,4-oxadiazoles with polyfluoroarenes” showed the significant effect of copper catalysts in direct Csp2–H/Csp2–H coupling reactions under mild conditions. Previously, polyfluoroarene-containing 1,3,4-oxadiazoles were obtained by cyclization of 1-benzoyl-2-pentafluorobenzoylhydrazine. It was envisaged that it would be feasible to couple two C–H bonds to form this kind of biaryl compounds catalyzed by a copper catalyst. Acetonitrile as a solvent and an atmosphere of dioxygen played a key role in the reaction. A series of biaryl substrates were prepared in moderate to good yields. The third and fourth work “Transition metal-free direct C–H bond thiolation or phosphorylation of 1,3,4-oxadiazoles and related heteroarenes” exhibited the charm of transition metal-free reactions and provided new bond forming reactions for the functionalization of 1,3,4-oxadiazoles. In these two cases, the construction of C–S and C–P bonds was realized by the direct functionalization of a series of heteroarenes, respectively. In the former work, aside from 1,3,4-oxadiazoles, indoles, benzothiazole, N-phenylbenzimidazole and caffeine were also thiolated to produce the corresponding products in excellent yields. In the latter work, a variety of 2-substituted 1,3,4-oxadiazoles and related heterocycles were phosphorylated in good to excellent yields. Furthermore, another reaction process was discovered involving copper-catalyzed reactions “Copper-catalyzed synthesis of alpha-thiophenyl carbonyl compounds through S–S bond cleavage and C–C bond activation”, in which a new procedure for the synthesis of alpha-thiophenyl carbonyl compounds through copper-catalyzed dual C–C/S–S bond cleavages was reported. Despite a variety of carbon-carbon bond formations by selective C–C cleavages, however, it was the first time to form C–S bonds through selective C–C cleavage of beta-diketones. Acetonitrile was crucial for the success of the reaction. Under the optimized reaction conditions, a series of alpha-arylthioketones and alpha-arylthioesters were synthesized from diaryl disulfides and beta-diketones or beta-ketoesters. Both alkyl and aryl functionalized beta-diketones/beta-ketoesters, as well as a variety of diaryl disulfides were well applied in the reaction.

Im Rahmen dieser Dissertation wurden Kupfer-katalysierte Reaktionen und deren Anwendung in der organischen Synthese untersucht. Hierbei wurden neue Methoden zur direkten C-C-, C-S- und C-P-Bindungsknüpfung von Heterozyklen entwickelt, von denen zwei unter metallfreien Reaktionsbedingungen ablaufen. Im ersten Abschnitt „The copper-catalyzed Sonogashira-Hagihara-type cross-coupling reactions“ werden Kreuzkupplungen von Arylhalogeniden mit terminalen Alkinen beschrieben, die durch sub-mol% Mengen an Kupfersalzen katalysiert werden. Im Laufe der Untersuchungen stellten sich alle getesteten Kupfersalze als katalytisch aktiv heraus, und es konnte gezeigt werden, dass die Struktur des Liganden für die erfolgreiche Durchführung der Reaktion entscheidend war. Zusätzlich wurde die Substratbreite und mechanistische Details mittels Kinetikmessungen und DFT (Dichtefunktionaltheorie) untersucht. Im zweiten Forschungsprojekt „Copper-mediated direct oxidative cross-couplings of 1,3,4-oxadiazoles with polyfluoroarenes“ wurde der entscheidende Einfluss von Kupferkatalysatoren auf direkte nach milden Reaktionsbedingungen ablaufende Csp2-H/Csp2-H Kupplungsreaktionen untersucht. Bisher wurden Polyfluoraren-haltige 1,3,4-Oxadiazole durch Zyklisierung von 1-Benzoyl-2-pentafluorobenzoylhydrazine erhalten. Es wurde gezeigt, dass die Bindungsbildung zwischen zwei C-H-Bindungen unter Bildung einer biarylischen Verbindung mit Hilfe eines Kupferkatalysators möglich ist. Hierbei waren Acetonitril als Lösungsmittel und reiner Sauerstoff für den Ablauf der Reaktion wichtig. Eine Vielzahl von biarylischen Substraten konnte in moderaten bis guten Ausbeuten erhalten werden. Der folgende Abschnitt, mit dem Titel „Transition metal-free direct C-H bond thiolation or phosphorylation of 1,3,4-oxadiazoles and related heteroarenes“, lag die Idee zugrunde, neue Bindungsbildungsreaktionen für die Funktionalisierung von 1,3,4-Oxadiazolen unter übergangsmetallfreien Reaktionsbedingungen zu ermöglichen. Es konnte die C-S bzw. die C-P Bindungsbildung über eine direkte Funktionalisierung von einer Reihe von Heteroarenen ermöglicht werden. Im Speziellen wurden im dritten Forschungsprojekt neben 1,3,4-Oxadiazolen auch Indole, Benzothiazole, N-Phenylbenzimidazol und Koffein thioliert. Die entsprechenden Produkte ließen sich in exzellenten Ausbeuten erhalten. Weiterhin konnte im vierten Forschungsprojekt eine Vielzahl von 2-substituierten Oxadiazolen und verwandte Heterozyklen phosphoryliert und in guten bis exzellenten Ausbeuten erhalten werden. Darüberhinaus wurde eine weitere Kupfer-katalysierte Reaktion „Copper-catalyzed synthesis of alpha-thiophenyl carbonyl compounds through S-S bond cleavage and C-C bond activation“ entdeckt, mit der alpha-Thiophenyl-carbonylverbindungen durch einen Kupfer-katalysierten dualen C-C/S-S-Bindungsbruch entstehen. Trotz einer Vielzahl von C-C- Bindungsbildungsreaktionen durch selektiven C-C-Bindungsbruch, war dies das erste Beispiel für die Bildung einer C-S-Bindung durch selektiven C-C-Bindungsbruch von beta-Diketonen. Acetonitril war erneut entscheidend für den Erfolg der Reaktion. Mit den optimierten Reaktionsbedingungen konnten aus den entsprechenden Diarylsulfiden, beta-Diketonen und beta-Ketoestern verschiedene alpha-Arylthioketone und alpha-Arylthioester synthetisiert werden. Sowohl Alkyl- als auch Aryl- funktionalisierte beta-Diketone/beta-Ketoester, wie auch eine Vielzahl von Diaryldisulfiden ließen sich in der Reaktion umsetzen.
