Kurzfassung
Im Gegensatz zu Sulfon-basierten Arzneistoffen sind Sulfoximin- und Sulfilimin-basierte Medikamente bis heute weniger bekannt. Mit der Zielsetzung die Anzahl der Sulfoximin-basierten bioaktiven Verbindungen zu erweitern, wurden verschiedene potentiell bioaktive Derivate, wie beispielsweise azyklische Triarylolefine, Sulfoximin- und Sulfilimin-basierte Dapson-Analoga und Sulfonimidoylfluoride synthetisiert, und deren bioaktive Eigenschaften untersucht. Es zeigte sich, dass Sulfoximin-basierte azyklische Triarylolefine ohne ER-Selektivität an Östrogenrezeptoren binden und keinerlei COX-Inhibition zeigen. Zudem zeigen weder Sulfoximin- noch Sulfilimin-basierte Dapson-Analoga COX-Inhibition und keinerlei Interaktion mit LTB4 und TNF. Außerdem wurden neue kupferkatalysierte Methoden der N-Alkinylierung von NH-Sulfoximinen mit Bromalkinen entwickelt. Eine Vielzahl neuartiger N-alkinsubstituierter Sulfoximine wurde in guten bis zu exzellenten Ausbeuten unter milden basischen Reaktionsbedingungen erhalten.Compared to sulfone-based drugs, sulfoximine- and sulfilimine-based bioactive compounds are not so common. In order to enlarge the number of sulfoximine-based bioactive compounds, several new potential bioactive sulfonimidoyl derivatives, including bioactive sulfoximine-based acyclic triaryl olefins, sulfoximine- and sulfilimine-based dapsone analogues and bioactive sulfonimidoyl fluorides have been synthesized, and some of their biological properties have been investigated. The sulfoximine-based acyclic triaryl olefins were good binding to the estrogen receptors ,but without ER-selectivity. Unfortunately, they also showed no COX inhibition. Furthermore, neither the sulfoximine- nor the sulfilimine-based dapsone analogues showed COX inhibition, and both did not interact with LTB4 and TNF. In addition, initial studies of the effect induced by O/N-exchange of sulfoximine-based bioactive compounds were conducted. A new methodology for the N-alkynylation of sulfoximines using N-H sulfoximines and bromoacetylenes catalyzed by inexpensive copper salts was developed. A variety of N-alkynylated sulfoximines was obtained in good to excellent yields under mild base conditions.
