Kurzfassung
Die Thematik der vorliegenden Dissertation war die Spezifizierung der kovalenten posttranslationalen Modifikation membranständiger Proteine, in diesem Falle des Glyzinrezeptors, durch Ubiquitinierung und Sumoylierung, um so die Mechanismen, die zur Internalisierung des Rezeptors führen, darzustellen. Die klinische Relevanz dieser Aspekte ist weitläufig und besteht in diesem Zusammenhang vornehmlich in der Fehlregulation der Rezeptorexpression und den jeweils zu erwartenden Konsequenzen. Um Ubiquitinierung bzw. Sumoylierung nachzuweisen wurden Versuche durchgeführt, in denen speziell zubereitete Lysate aus Ratten-Gehirn- und Rückenmarksgewebe mit Sumo bzw. Ubiquitin und verschiedenen Glyzinrezeptor-Domänen und -Mutanten inkubiert wurden. Die Proteine wurden als GST-Fusionsproteine rekombinant aus E. coli gereinigt und oder duch Expression in Xenopus-laevis-Oocyten radioaktiv markiert und waren durch GST- oder Hexahistidylsequenzen isolierbar. In dieser Art von Versuchen ließen sich keinerlei Addukte zwischen dem GlyR und Ubiquitin oder Sumo nachweisen. Zusätzlich wurden Experimente zur Ubiquitinierung und Sumoylierung an intakten Xenopus-Oozyten durchgeführt, in die hierzu geeignete Proteine und cRNA injiziert wurden. Nach [35S]Methioninmarkierung wurden die exprimierten Proteine durch GST-Pulldown und Ni2+-NTA-Chelatchromatografie aufgereinigt. Mittels dieser Methodik konnte eine Monoubiquitinierung des Aöpha 1-Glyzinrezeptors dargestellt werden. Eine Glyzinrezeptor-Mutante ohne zytoplasmatische Lysine, Alpha 1 NoK-GlyR, wurde nicht ubiquitiniert, zeigte allerdings die gleichen proteolytischen Spaltprodukte wie der Wildtyp Alpha 1-Glyzinrezeptor. Dies impliziert, dass es neben einem Ubiquitin-abhängigen Weg noch einen parallelen Mechanismus geben muss, der die Internalisierung des Glyzinrezeptors mit anschließendem lysozymalen Abbau in Xenopus-laevis-Oozyten vermittelt. Zudem zeigte sich, dass der Wildtyp Glyzinrezeptor ebenso wie die ubiquitinierungssdefiziente Glyzinrezeptor-Mutante Alpha 1NoK-GlyR eine nicht-kovalente Bindung mit Ubiquitin eingeht. Es ist möglich, dass eine solche nicht-kovalente Anlagerung der kovalente Modifizierung vorausgeht. Interessant verhielt sich zudem die Glyzinrezeptor-Mutante I387A,D388A-Alpha 1, die ubiquitiniert wurde, ohne an der Zellmembran exprimiert zu werden. Möglicherweise dient die Ubiqutinierung in diesem Falle der proteasomalen Degradation, da diese Mutante nicht in den Golgi-Apparat und spätere Kompartimente exportiert wurde. Die Endodomäne des Glyzinrezeptors, die die große zytoplasmatische Schleife zwischen den Transmembrandomänen 3 und 4 und damit alle 10 potentiellen Ubiquitinierungslysine umfasst, wurde in den Oozyten nicht ubiquitiniert. Spekulativ ist anzunehmen, dass die entsprechenden Ubiquitinierungsenzyme nur den membranständigen Glyzinrezeptor, nicht aber die lösliche Endodomäne als Substrat erkennen. Eine GlyR-Sumoylierung ließ sich weder in Oozyten noch in-vitro unter Mithilfe der Ubc09-Ligase detektieren. Allerdings fiel in Sumo-überexprimierenden Oozyten eine Zunahme der für die Endozytose des Glyzinrezeptors typischen proteolytischen Spaltung auf. Da dieser Effekt des Sumos unabhängig von der Anzahl der Lysine des Rezeptors stattfand, wie sich durch verschiedene Mutanten zeigen ließ, ist anzunehmen, dass die Sumo-Wirkung indirekt und nicht durch direkte Sumoylierung zustande kommt.The specification of covalent posttranslational modification by sumoylation and ubiquitination of membrane proteins – in this case of the GlyR alpha 1-subunit and its ubiquitin mediated internalisation – is described. The clinical relevance of those mechanisms is wide spread and would in this case produce an inadequate receptor regulation with the expected consequences. In order to show ubiquitination/sumoylation lysates of rat brain and spinal cord were incubated with ubiquitin/sumo as well as different mutants of the glycine subunit. The proteins were isolated as GST-fusion-protein from E. coli or were expressed as radioactive His-tagged protein in Xenopus oocytes. After incubation it was expected that adducts of ubiquitin/sumo with the receptor could be isolated by using GST-pullover or Ni2+-NTA-Chromatographie. However, there were no adducts found. In further experiments the ubiquitination/sumoylation of the receptor was demonstrated in Xenopus oocytes, in which cRNA and Proteins of the different components were injected before incubation with radioactive [S]-methionine. The isolation of adducts was done by GST-pullover or Ni2+-NTA-Chromatographie; the adducts were demonstrated on SDS-PAGES. It was therefore possible to show monoubiquitination of the GlyR alpha 1-subunit. A special mutation without cytoplasmatic lysines was not ubiquitinated, however was degraded into the same proteolytic pieces specific for ubiquitin mediated endocytosis as the wildtype. This implies that there is an alternate way to mediate endocytosis rather than ubiquitination. Further more a non covalent binding of ubiquitin to the wildtype and to the No-K-mutant was observed implicating a non covalent binding before covalent modification. Interestingly, a special mutant I387A,D388A GlyR alpha 1-subunit was ubiquitinated without being expressed at the cell membrane. In this case it might be ubiquitination serves in this case to address the mutant, which was never transported to the golgi, for proteasomal degradation. The endodomain of the receptor could be excluded as substrate of ubiquitination. We supposed that the ubiquitination enzymes cannot recognize the soluble endodomain, they may need it fixed at the membrane. A sumoylation of the GlyR alpha 1-subunit could not be detected even in ubc-09-ligase over expressed cells. But in the sumo expressing oocytes was an increased rate of receptor endocytosis. This effect was independent of the lysins in the Protein, so we suppose an indirect effect or a non-covalent binding.
