Kurzfassung
Sulfoximine spielen eine wichtige Rolle in den pharmazeutischen und agrochemischen Wissenschaften. Sie sind wichtige Bausteine von Pseudopeptiden und bilden wirkungsvolle Liganden in der asymmetrischen Katalyse. Es ist daher notwendig eine einfache, effiziente und skalierbare Methode zu entwickeln, die einen leichten Zugang zu Sulfoximinen oder ihren Derivaten ermöglicht. Die Darstellung erfolgt entweder durch Übergangsmetallkatalyse oder metallfreie Iminierung. Diese Arbeit beschreibt ein metallfreies Scale-up-Verfahren zur Herstellung von N-Cyanosulfiliminen und ihre anschließende Oxidation zu N-Cyanosulfoximinen. Weiterhin sind zwei Methoden zur Entschützung eines N-Cyanomethylphenylsulfoximines zu einem synthetisch wertvollen NH-Sulfoximin im Grammmaßstab beschrieben. Darüber hinaus wird ein Verfahren zur Gewinnung eines heterocyclischen Sulfoximins, ausgehend von einem Dialkylsulfoximin vorgestellt. Außerdem konnte die Synthese eines chiralen, nicht racemischen Sulfoximin analogos von Myristinsäure in guten Ausbeuten und mit hohem Enantiomerenüberschuss (>99% ee) unter Verwendung eines asymmetrischen Oxidationsverfahrens und einer Methode zur Eisenkatalysierten Iminierung konnte detailliert beschrieben werden.

Sulfoximines play an important role in the pharmaceutical and agrochemical sciences and are noteworthy as building blocks of pseudopeptides and as ligands in asymmetric catalysis. Hence, it becomes imperative to have a simple, efficient and scalable method which can provide easy access to sulfoximines or their derivatives. Sulfoximines can be synthesized by either a transition metal catalysed or a metal-free imination method. This thesis describes a metal-free, scale-up procedure for the preparation of N-cyano sulfilimines and their subsequent oxidation to form N-cyano sulfoximines. Furthermore, two approaches for the gram-scale deprotection of an N-cyano methyl phenyl sulfoximine to obtain a synthetically valuable NH-sulfoximine are also disclosed. In addition, a process for obtaining a heterocyclic sulfoximine, starting from a dialkyl sulfoximine, is reported. The synthesis of a chiral, non-racemic sulfoximine analogue of myristic acid, utilizing an asymmetric oxidation technique and an iron-catalyzed imination method, with good yields and high enantiomeric ratio (>99% ee), is also detailed herein.
